FEATURES OF HAEMATOLOGICAL PARAMETER CHANGES IN C57BL/6 MICE DURING PRIMARY TUMOR GROWTH AND METASTATIC PROGRESSION OF LEWIS LUNG CARCINOMA

It is well known that tremendous progress has been made to date in understanding the pathogenesis of malignant tumours and developing effective treatments that have significantly improved the prognosis for cancer patients. However, challenges remain in detecting metastatic spread before overt metastases are established and in predicting which patients are at most significant risk of developing metastatic disease, which is the leading cause of death. Screening tests can help to improve the management of metastatic disease, even before symptoms appear, by providing an accurate picture of when metastatic spread occurs and how this process determines the risk profile of cancer patients at diagnosis, throughout treatment, and in remission. One of the most reliable paraclinical methods of diagnosing diseases is the assessment of haematological blood parameters, which allows the detection of the progression of the oncological process. Today, special attention is focused on disorders in the hemostatic system, which are one of the most essential components of the pathogenesis of the tumour process. This study aimed to evaluate the hemogram parameters of male C57BL/6 mice with primary Lewis lung carcinoma (LL) tumour and its metastatic progression and assess changes under the conditions of direct-acting anticoagulants to determine biomarkers associated with metastasis. Based on the studies, a significant decrease in the total platelet count and thrombocrit percentage was found against the background of growth of the primary LL carcinoma tumor and the background of its metastatic progression. Regarding the platelet index – the relative width of platelet distribution by volume (an indicator of platelet heterogeneity), a correlation with the progression of LL carcinoma metastasis was noted. This indicator increased by 15.21-41.65% in the background of the primary tumour – there were no differences compared to intact animals. The use of direct-acting anticoagulants enoxaparin and sulodexide, which inhibited the formation of metastases in the lungs of animals by 60.88% and 63.79%, respectively, significantly increased the number of platelets and decreased the percentage of relative platelet distribution by volume by 15.21% and 1.8%, respectively.
