The search for simple, non-invasive and non-toxic methods of visualizing neural cells as part of three-dimensional structures is an urgent problem in cell biology and biotechnology. As such a method, in vivo autofluorescence can be used to provide information about the viability, morphological, and physiological state of neural cells in three-dimensional structures. The aim is to investigate the possibility of using NAD(P)H and flavin autofluorescence to visualize and assess the viability of neuronal cells within aggregates and spheroids. In the experiments on three-dimensional aggregates and spheroids of neural cells obtained from the brain tissue of newborn rats, the autofluorescence of cells was studied depending on their viability. It was found that the use of a laser with a wavelength of 405 nm leads to the excitation of neural cell autofluorescence in the green region of the spectrum, which allows for non-invasive, in vivo visualization of living cells as part of three-dimensional structures and estimation of their number and packing density. Analyzing the intensity of green autofluorescence of neural cells in three-dimensional structures opens opportunities for monitoring their metabolic state in real-time.
