The overall aim of the investigation is to study the reconstruction of the hemomicrocirculating vessel bed of the white rat tongue throughout different stages of experimental diabetes mellitus. Thus the present study has investigated the rat tongue on the 2nd, 4th, 6th and 8th weeks of experimental streptozotocin-induced diabetes. Male white pubertal rats of 130-150 g in weight were used in the experiments and divided into 5 exercise groups. All the experiments were approved by the University Animal Care and Use Bioethical Committee (Protocol #1/2009). During the investigation, the anatomical, injectional, morphometric and biochemical methods were used. A morphometrical analysis of the white rat hemomicrocirculating vessel bed throughout different periods of streptozotocin-induced diabetes was carried out. It was established that numerous dorsal arterial branches run from the deep lingual artery. Those dorsal branches are divided into vascular networks of the mucous membrane of the proper lamina. The morphometrical parameters of the hemomicrocirculating vessel bed of the white rat tongue are the following: the diameter of arteriolas equal – 25.50±0,10 µm; of capillaries – 5.10±0.06 µm; and of venulas – 38.5±0.11 µm. The arteriolar-venular coefficient equals 0.670±0.003; the density of exchange vessels are as follows: for arteriolas – 0.080±0.001; for capillaries – 0.150±0.001, and for venulas – 0.035±0.001. The vascular sinuousity coefficient equals 0.960±0.002. The investigation of the morphological changes started after two weeks of streptozotocin-induced diabetes. The first changes in the hemomicrocirculating vessel bed were marked, especially the enlargement of the deep lingual artery lumen and dorsal branches of the tongue. The lumen of arteriolas was irregular in shape, whereas the lumen of venulas was partly constricted. The mucous membrane capillary networks of the lingual dorsum was preserved. After four weeks of streptozotocin-induced diabetes, most arteriolas and capillaries were spasmodic. The great vessels of the tongue were constricted (i.e. the deep artery of the tongue and its dorsal branches). The capillaries of the tongue were constricted and lost their lace-like patterns. At the end of the sixth week of experimental streptozotocin-induced diabetes, the destructive changes in the hemomicrocirculating vessel bed were marked with simultaneous compensatory changes. The arteria-arterial, arteriola-arteriola and arteriola-venula anastomoses joined the process. All the morphometrical parameters of the hemomicrocirculating vessel bed of the white rat tongue changed after the sixth week of the experimental diabetes mellitus. Our data show that after the eighth week of streptozotocin-induced diabetes the following deep changes in the hemomicrocirculating vessel bed of the rat tongue were observed: the enlargement of every vascular component, the destruction of capillary components and the thickening and sclerosis of vascular walls. The capillary networks were destroyed. These processes were combined with the enlargement of arteriolar-venular anastomoses. The blood directly ran to the dilated venular component. Big vessels were enlarged in the region of the tongue root. The following deep structural changes in the hemomicrocirculating vessel bed of the rat tongue have been established: diameters of vessels increased (arteriolas – to 28.60±0.11 µm; precapillary arteriolas – to 13.30±0,09 µm; capillaries to – 5.9±0.07 µm; postcapillary venulas to – 14.20±0.09 µm; venulas to – 45.30±0.15 µm). The arteriolar-venular coefficient decreased to 0.631±0.003; the density of the exchange vessels, such as arteriolas and capillaries, decreased to 0.060±0.007 and 0.10±0.01; venulas increased to – 0.025±0.001. The vascular sinuousity coefficient decreased to 0.575±0.008. Having analyzed of the morphometric parameters, we are able to confirm that the pathological process after eight weeks of streptozotocin-induced diabetes becomes irreversible. The dilated vessels (arteriolas) and sclerosal and thickened walls of the hemomicrocirculating vessel bed do not provide adequate blood the tongue.
