Antimicrobial resistance has led to growing interest in bacteriophages as an adjunct to antibiotics therapy. The impact of cultivation conditions on phage reproduction remains insufficiently studied. This study evaluates the effect of glucose and calcium ions (Ca²⁺) on the ability of bacteriophages to lyse bacterial cultures. Bacteriophages used in the study were isolated from urban wastewater in Kyiv and belonged to the morphotypes Siphoviridae, Podoviridae, and Myoviridae. The effect of glucose (1%) and calcium ions (Ca²⁺, 50 mM) on plaque formation was studied by incorporating these factors into nutrient agar for microbial cultivation. The results were assessed by comparing plaque numbers on media with and without the added factors. The experiment demonstrated that glucose and Ca²⁺ ions had different effects on plaque formation in various bacteriophages. Glucose often inhibited phage activity, whereas Ca²⁺ ions generally enhanced plaque formation (either by increasing plaque number or size). In some cases, glucose also promoted phage reproduction. The influence of different factors on phage reproduction should be considered when isolating and characterizing bacteriophages, as well as in the development of phage therapy preparations.
