GENOME AVAILABILITY: CHANGES IN THE CHROMATIN OF CELLS OF THE FOLLICULAR ENVIRONMENT OF OOCYTES UNDER CONDITIONS OF OXIDATIVE STRESS IN VITRO

In this work, new data on the effect of H2O2 (5 min, 250 mM) on the cells of the follicular environment of oocytes were obtained, which give grounds to assert that such changes in the values of cytological parameters, namely a decrease in the chromatinization index (ChI) as a decrease in genome activity due to a decrease in its accessibility ("openness"), an increase in the nucleolar index (NI) as an increase in the activity of the translational apparatus - the reaction of cells to stress, an increase in the index of morphologically altered nuclei (MNI) as a decrease in the integrity of the karyoplasm - the functioning of the genome is suppressed, together reflect chromatin damage characteristic of oxidative stress. The same changes are observed under the influence of a PARP (poly(ADP-ribose) polymerase) blocker and an iNOS blocker. The use of NOS substrate and SIRT-1 activator, under the influence of a PARP blocker, an iNOS blocker and, together, a PARP blocker and an iNOS blocker, led to: an increase in ChI - as an increase in genome activity due to an increase in its accessibility ("openness"), a decrease in NI - as a decrease in the activity of the translational apparatus - a decrease in the cellular response to stress, a decrease in MNI - as an increase in the integrity of the karyoplasm - the functioning of the genome is restored, together reflecting the restoration of chromatin activity. The data obtained suggest that nitric oxide and SIRT-1 are involved in maintaining the functional state of chromatin availability in the cells of the follicular environment of oocytes.
