Causes of mitotic disorders in hepatocytes include toxic substances, viral infections, genetic mutations, inflammatory stress, and nutritional deficiencies. Mitotic dysfunction can lead to severe pathologies such as hepatocellular carcinoma, fibrosis, cirrhosis, and liver failure. Genomic instability may impair the liver's regenerative capacity, further deteriorating its functional state. This study aimed to determine the distribution of the ploidy index, nuclear index, and mitotic number in hepatocyte subtypes under conditions of central blocking of luteinizing hormone synthesis with long-term exposure to tryptorelin and dietary quercetin supplementation in animals. The experiment was conducted on 60 sexually mature male white rats weighing 140–160 g. Liver tissue was used as the study material. The animals were divided into three groups. Group 1, the control group, received saline (10 animals). Group 2 was subcutaneously injected with diphereline (tryptorelin embonate) at a dose of 0.3 mg of active ingredient per kg of body weight for a duration of 365 days (25 animals). Group 3 was administered a solution of tryptorelin at the same dose, with quercetin added to the diet through a gastric tube, based on body weight, three times a week (25 animals). The animals were euthanized at 1, 3, 6, 9, and 12 months with an overdose of ether anesthesia. Prolonged central blocking of the synthesis of luteinizing hormone by tryptorelin leads to oxidative damage to the liver parenchyma. The reaction of the liver parenchyma was manifested by an increase in the ploidy index, nuclear index and mitotic number in hepatocytes at later stages of the study. Quercetin positively affects the course of oxidative stress in the liver.
