Ultraviolet radiation is a natural stimulator of melanogenesis and a key factor in hyperpigmentation. It changes the microenvironment of tissues, stimulating melanin formation. At the same time, UV radiation is used in the treatment of certain skin diseases, but it can cause unwanted hyperpigmentation disorders, for the pharmacotherapy of which hydroquinone, a tyrosinase inhibitor, is used in practice. The aim is to study the skin's reaction to UV irradiation and skin readaptation without intervention and with the use of hydroquinone. Rats with pigmented fur were depilated in the interscapular region, after which they were subjected to subery- themal UV irradiation (311 nm) 15 times over 30 days. The animals were divided into two groups: a free readaptation group and a hydroquinone depigmentation group. Skin biopsies were performed on days 31, 45, 61, and 121 of the experiment. The studies included staining with hematoxylin-eosin and the Fontana-Masson method. Morphometric analysis was performed using ImageJ software. Additionally, a synthetic “opacity index” (OI) was calculated as the product of epidermal thickness and density of melanosomes (MS). UV irradiation increased the thickness of the epidermis and the percentage of the epidermal cross-sectional area occupied by melanosomes on day 31 (OI = 264.3%). The OI value in the free readaptation group gradually decreased to 163.6%, and in the hydroquinone group to 159.7%. The OI was lower in the hydroquinone group on days 45 and 61. UV radiation increases the thickness of the epidermis and the density of melanosomes. OI remains elevated at all stages of the experiment. Hydroquinone effectively reduces the opacity index.
