COMPARATIVE ANALYSIS OF BIOCHEMICAL PROFILES OF HUMAN PLACENTAL EXPLANTS BY FETAL SEX AFTER CRYOPRESERVATION

Preservation of cell viability and morphological integrity of placental explants is crucial for their subsequent use in pharmacology and regenerative medicine. Of particular interest is the potential influence of fetal sex on the bio chemical profile of the placenta. The aim of this study was to determine the efficiency of cryopreservation of human placental explants using 10% dimethyl sulfoxide (DMSO) and to assess the effect of fetal sex on the content of biologically active substances in placental extracts. Placental explants were obtained from women after cesarean section and cryopreserved at a cooling rate of 1°C/ min to –70°C, followed by storage in liquid nitrogen. Cell viability was assessed using vital staining with neutral red and trypan blue, while morphological analysis was performed after hematoxylin and eosin staining of histological sections. Hormone and protein levels were determined using electrochemiluminescence and photometric methods. After cryopreservation with DMSO, high cell viability and preservation of placental villous morphological integrity were observed, with minimal transient signs of damage. No morphological differences were detected between sam ples obtained from fetuses of different sexes. Biochemical analysis revealed sex-specific differences: extracts derived from placental tissue of female fetuses contained higher levels of alpha-fetoprotein, chorionic gonadotropin, lutein izing hormone, prolactin, and total protein, whereas samples from male fetuses were characterized by increased levels of testosterone and unconjugated estriol. Thus, cryopreservation with DMSO ensures effective preservation of the structural and functional integrity of placental explants. At the same time, fetal sex determines the specific biochemical composition of the tissue, which is important for the further development of placental-based preparations for medical use.
